Analysis of Ion Channels in Native Tissues

Like other permeation pathways of biofilms, ion channels are essential to life because they generate action potentials and regulate synaptic transmission in neurons and muscle cells, underlie intracellular Ca2+ signaling, and contribute to separate charges across the plasma membrane. Gene defects or acute dysregulation of ion channel activity, trafficking, or degradation often contribute to severe channelopathies and pathologies.

Due to the heteromultimeric co-assembly of channel α-subunits with other β-subunits, other interacting subunits, and other modifications (such as glycosylation, alternative splicing of mRNA, and phosphorylation), the number of channels with different functions in natural tissues is further increased. There is a need to utilize multiple advanced techniques to understand the properties and functions of ion channels present in natural tissues to accelerate the development of drugs targeting ion channels.
